Biological Background: ICAM-1/CD54 Function and Localization

  • Functions as a receptor ligand for ITGAL:ITGB2/LFA-1, enabling leukocyte cell-cell adhesion and providing a calibrated system to adjust T-cell killing strength based on antigen stimulation. Related references: PMID:3086451, PMID:3340213, PMID:38195629
  • Also acts as a ligand for ITGAM:ITGB2/MAC-1, mediating adhesion between stimulated neutrophils and stimulated endothelial cells. Related references: PMID:1980124
  • During leukocyte trans-endothelial migration, ICAM1 engagement promotes the assembly of endothelial apical cups through ARHGEF26/SGEF and RHOG activation. Related references: PMID:17875742
  • Promotes cell aggregation in epithelial cells via interaction with MUC1. Related references: PMID:11173916
  • Acts as a receptor for major group rhinovirus A and B capsid proteins, as well as Coxsackievirus A21 capsid proteins, facilitating viral entry.
  • Targeted for degradation by the Kaposi's sarcoma-associated herpesvirus/HHV-8 E3 ubiquitin ligase MIR2, presumably to evade immune cell lysis.
  • Localizes to the cell membrane and is expressed on non-hematopoietic cells (vascular endothelium, thymic epithelium, fibroblasts) and hematopoietic cells (tissue macrophages, stimulated T lymphoblasts, dendritic cells). Expressed at low levels on peripheral blood leukocytes. Related references: PMID:3086451
  • Undergoes post-translational modifications including disulfide bond formation, N-linked glycosylation, and phosphorylation; also subject to ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 28–480 of the extracellular domain, ensuring recognition of native surface ICAM-1.
  • This conjugate is optimized for flow cytometry; titrate the antibody in your specific assay system to determine the optimal working concentration.
  • For best results, use human cell lines or primary cells known to express ICAM-1 (e.g., HUVECs, Jurkat T cells stimulated with mitogen).
  • Avoid repeated freeze-thaw cycles; store according to shipping conditions (ice bag) upon receipt.
