  • Isoleucine--tRNA ligase, cytoplasmic (IARS) is a member of the class I aminoacyl-tRNA synthetase family, also known as isoleucyl-tRNA synthetase (IRS) or IleRS.
  • Catalyzes the specific attachment of isoleucine to its cognate tRNA(Ile) in a two-step reaction: activation of isoleucine by ATP to form Ile-AMP, followed by transfer to the tRNA acceptor end, a critical step in protein biosynthesis.
  • Encoded by the IARS1 gene (GeneID: 3376) in humans.
  • Primarily localized in the cytoplasm and cytosol.
  • Subject to post-translational modifications including acetylation and phosphorylation (as identified by proteomics).
  • Clinically, mutations in IARS1 have been linked to intellectual disability (disease variant).
  • Expressed at the protein level in liver and muscle tissues.
