Biological Background: Isoleucine--tRNA ligase, cytoplasmic Function and Localization
- Isoleucine--tRNA ligase, cytoplasmic (IARS) is a member of the class I aminoacyl-tRNA synthetase family, also known as isoleucyl-tRNA synthetase (IRS) or IleRS.
- Catalyzes the specific attachment of isoleucine to its cognate tRNA(Ile) in a two-step reaction: activation of isoleucine by ATP to form Ile-AMP, followed by transfer to the tRNA acceptor end, a critical step in protein biosynthesis.
- Encoded by the IARS1 gene (GeneID: 3376) in humans.
- Primarily localized in the cytoplasm and cytosol.
- Subject to post-translational modifications including acetylation and phosphorylation (as identified by proteomics).
- Clinically, mutations in IARS1 have been linked to intellectual disability (disease variant).
- Expressed at the protein level in liver and muscle tissues.
- Predicted molecular weight is ~144.5 kDa (UniProt P41252), consistent with the observed band at 144 kDa.
