Biological Background: FATP1 Function and Localization

  • FATP1 (SLC27A1), also known as ACSVL5, FATP-1, long-chain fatty acid transport protein 1, arachidonate--CoA ligase, and very long-chain acyl-CoA synthetase, is a member of the solute carrier family 27. It is subject to alternative splicing, yielding multiple isoforms.
  • Core function 1: Mediates cellular import of long-chain fatty acids (LCFA) at the plasma membrane.

  • Core function 2: Also acts as an acyl-CoA ligase, catalyzing ATP-dependent formation of fatty acyl-CoA from LCFA and very-long-chain fatty acids (VLCFA), preventing efflux and promoting net fatty acid accumulation.
  • Subcellular localization: Localizes to the cell membrane and endomembrane system, and is present in the cytoplasm, consistent with roles in transport and intracellular fatty acid activation.
  • Tissue distribution: Highest expression in muscle and adipose tissue, moderate in small intestine, very low in liver; also detected in brain gray matter.
