Biological Background: CD98 (SLC3A2) Function and Localization
- CD98/SLC3A2, also known as 4F2 heavy chain, is a type II transmembrane glycoprotein that serves as the heavy chain subunit of the 4F2 cell-surface antigen. Alternative splicing produces multiple isoforms with varying molecular weights.
- It functions as a chaperone for biogenesis and trafficking of amino acid transporters. It forms heterodimers with various SLC7 family light chains, including SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A10, and SLC7A11, to facilitate the exchange of neutral, dibasic, and anionic amino acids across the plasma membrane (PubMed:10574970, PubMed:10903140, PubMed:11557028, PubMed:34880232, PubMed:9829974, PubMed:11389679). Substrate specificity is determined by the SLC7A subunit; for example, SLC3A2/SLC7A11 mediates cystine/glutamate antiport.

- SLC3A2 is indispensable for the plasma membrane localization, stability, and transport activity of its heterodimeric partners (PubMed:10391915, PubMed:10574970, PubMed:11311135). Interaction with LAPTM4B redirects the SLC3A2/SLC7A5 complex to lysosomes, stimulating mTORC1 signaling via leucine uptake (PubMed:25998567).
- Related references: PMID:10391915 PMID:10574970 PMID:11311135 PMID:25998567
- Beyond transport, CD98 modulates integrin-dependent signaling, contributing to cell spreading, migration, and tumor progression (PubMed:11121428, PubMed:15625115).
- Related references: PMID:11121428 PMID:15625115
- CD98 is expressed at the cell membrane, apical and basolateral surfaces, and is also found in lysosomes, melanosomes, and cell junctions.
- The protein undergoes various post-translational modifications including acetylation, phosphorylation, disulfide bond formation, and ubiquitination.
- Widely expressed, CD98 shows highest levels in kidney, placenta, and testis, with lower expression in thymus and resting T cells. It also mediates entry for hepatitis C virus and serves as a receptor for Plasmodium vivax in reticulocytes.
Experimental Guidance and Technical Tips
- As this antibody is directly conjugated to ABflo® 594, it is ready for flow cytometry without secondary staining. Consider titrating to determine the optimal concentration for your specific cell type.
- Validate staining in the appropriate sample system (e.g., human peripheral blood mononuclear cells or cell lines known to express CD98).
- Use an isotype control matched to the host species and conjugate to assess nonspecific binding.
- For multicolor panels, confirm that ABflo® 594 is compatible with your instrument’s configuration and other fluorophores.
