Biological Background: CD82 Function and Localization
- CD82, also known as KAI1, C33, IA4, R2, and TSPAN27, is a member of the tetraspanin superfamily and functions as a metastasis suppressor.
- It is a structural component of tetraspanin-enriched microdomains (TERMs), which serve as platforms for receptor clustering and signaling. Related references: PMID:19497983
- Attenuates EGF signaling by facilitating EGFR endocytosis and desensitization, and regulates EGFR ubiquitination via CBL phosphorylation. Related references: PMID:10985391 PMID:35538033 PMID:23897813
- Modulates integrin-mediated cell adhesion and migration, including regulation of integrins alpha4, alpha6, and beta1. Related references: PMID:24623721 PMID:8757325 PMID:15557282 PMID:17560548
- Interferes with plasminogen activation by disrupting the PLAU-PLAUR interaction. Related references: PMID:15677461
- Plays roles in immune responses: costimulation of TCR/CD3 pathway, TLR9 trafficking (By similarity), and inhibition of LPS-induced TLR4 signaling. Related references: PMID:36945827
- Controls angiogenesis by binding and sequestering VEGFA and PDGFB, switching the angiogenic milieu to a quiescent state. Related references: PMID:34530889
- Subcellular localization: cell membrane, cytoplasmic vesicle, phagosome. Expressed in lymphoid tissues.
Experimental Guidance and Technical Tips
- For flow cytometry, consider titrating the antibody to determine the optimal staining concentration for your specific cell system.
- The immunogen corresponds to amino acids 1–267 of human CD82; verify that your target epitope is within this region, as some splice variants may not be recognized.
- Confirm cross-reactivity and specificity in your experimental setting, as this antibody is validated for human samples.
- Include appropriate isotype controls and fluorescence compensation when using this conjugated antibody in multicolor panels.
