Biological Background: Macrosialin (CD68) Function and Localization
- CD68 (macrosialin/gp110) is a heavily glycosylated type I transmembrane protein belonging to the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family.
- Plays a role in phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions.
- Binds to tissue- and organ-specific lectins or selectins, allowing homing of macrophage subsets to particular sites; rapid recirculation from endosomes/lysosomes to the plasma membrane may facilitate macrophage crawling.
- Highly expressed by blood monocytes and tissue macrophages; also found in lymphocytes, fibroblasts, endothelial cells, and many tumor cell lines, where it may mediate attachment to vascular endothelium and dissemination.
- Subcellular localization includes cell membrane, endosome membrane, and lysosome membrane, consistent with its role in endosomal/lysosomal trafficking.
