Biological Background: L-selectin Function and Localization

  • L-selectin (SELL; CD62L) is a calcium-dependent (C-type) lectin that mediates cell adhesion by binding to glycoprotein ligands on adjacent cells. Related references: PMID:12403782, PMID:28011641, PMID:28489325
  • It plays a critical role in lymphocyte homing by supporting the adherence of lymphocytes to high endothelial venules (HEV) of peripheral lymph nodes. Related references: PMID:12403782, PMID:28011641
  • L-selectin promotes the initial tethering and rolling of leukocytes along the vascular endothelium, a key step in the leukocyte adhesion cascade. Related references: PMID:12403782, PMID:28011641
  • The protein is a member of the selectin family of adhesion molecules (including E- and P-selectin) and contains an N-terminal C-type lectin domain, an EGF-like domain, multiple sushi (CCP) domains, a transmembrane helix, and a short cytoplasmic tail.
  • L-selectin is localized on the cell membrane and is constitutively expressed on most leukocytes, including B-cell lines and T-lymphocytes.
  • Following cellular activation, L-selectin is rapidly shed from the cell surface by proteolytic cleavage (a metalloprotease-mediated event), a process associated with its role in leukocyte migration.
  • The protein undergoes alternative initiation and is annotated with keywords including disulfide bond, glycoprotein, metal-binding, and proteomics identification.

Experimental Guidance and Technical Tips

  • As the immunogen corresponds to the N-terminal extracellular domains (aa 39–332), the antibody is expected to recognize the cell surface L-selectin, making it suitable for flow cytometry detection of live, unfixed cells.
  • Consider that L-selectin is shed from the cell surface upon activation; thus, staining protocols should minimize cell activation and be carefully timed. Validate in the relevant sample system.
  • The rabbit monoclonal format typically offers high specificity and batch-to-batch consistency; however, always confirm reactivity under the intended experimental conditions.
  • For flow cytometry, titrate the antibody to determine the optimal concentration and include appropriate isotype controls and viability dyes.
