Biological Background: CD59 Function and Localization

  • CD59, also known as protectin, MAC-inhibitory protein (MAC-IP), 20 kDa homologous restriction factor (HRF-20), and 1F5 antigen, is a GPI-anchored cell surface glycoprotein that functions as a critical inhibitor of the complement membrane attack complex (MAC).
  • It binds to the C8 component (C8A and C8B) of the assembling MAC, forming an intermolecular beta-sheet that prevents incorporation of multiple C9 molecules, thereby blocking formation of the osmolytic pore and protecting human cells from complement-mediated lysis (PubMed:11882685, PubMed:1698710, PubMed:2475111, PubMed:36797260).
