Biological Background: CD59 Function and Localization
- CD59, also known as protectin, MAC-inhibitory protein (MAC-IP), and 20 kDa homologous restriction factor (HRF20), is a GPI-anchored glycoprotein that serves as a critical complement regulator.

- CD59 is widely expressed on human cells, anchored to the plasma membrane via a glycosylphosphatidylinositol (GPI) moiety, and protects host cells from autologous complement-mediated lysis.
- A soluble form, found in urine, retains complement binding but exhibits greatly reduced MAC-inhibitory activity compared to the membrane-bound isoform.

- For flow cytometry, titrate the -conjugated antibody on human peripheral blood mononuclear cells (PBMCs) or CD59-expressing cell lines to determine optimal staining concentration.
- CD59 is GPI-anchored; avoid treatments that may release GPI-anchored proteins (e.g., phospholipase C) unless specifically evaluating anchor dependence. Validate in the relevant sample system.
