Biological Background: CD45RA Function and Localization
- CD45, encoded by PTPRC, is a receptor-type tyrosine-protein phosphatase essential for T-cell activation through the antigen receptor (PubMed:35767951). Related references: PMID:35767951
- Functions as a positive regulator of T-cell coactivation upon binding to DPP4.
- Contains two PTPase domains; the first has catalytic activity, while the second influences substrate specificity of the first.
- Recruits and dephosphorylates SKAP1 and FYN upon T-cell activation, and modulates LYN activity through dephosphorylation (By similarity).
- Interacts with CLEC10A on immature dendritic cells at the T-cell/APC contact site, raising the T-cell activation threshold to prevent autoreactivity.
- Acts as a receptor for human cytomegalovirus protein UL11, leading to reduced tyrosine phosphorylation and impaired T-cell proliferation upon TCR stimulation.
- Localizes to the cell membrane, membrane rafts, and immunological synapse, consistent with its role in signaling.
- Subject to extensive alternative splicing, generating multiple isoforms with distinct extracellular domains; the immunogen targets isoform 8 (CD45RA), which contains the RA exon.
- Heavily glycosylated and phosphorylated; disease-associated variants can cause severe combined immunodeficiency (SCID).

- The immunogen is a synthetic peptide from the N-terminal region (aa 10-110) of human CD45RA (isoform 8); perform sequence alignment to confirm specificity for the RA isoform and exclude cross-reactivity with other CD45 variants.
- For immunofluorescence on paraffin-embedded sections (IF-P), standard antigen retrieval protocol may be necessary; we recommend validating on human lymphoid tissue (e.g., tonsil, lymph node) as a positive control.
- Reported cross-reactivity is limited to human; if testing on other species, verify reactivity experimentally.
