Biological Background: CD355/CRTAM Function and Localization

  • CD355/CRTAM (Cytotoxic and regulatory T-cell molecule) is a member of the immunoglobulin superfamily that undergoes alternative splicing, yielding isoforms of approximately 44 kDa and 22 kDa.
  • It mediates heterophilic cell-cell adhesion that regulates the activation, differentiation, and tissue retention of various T-cell subsets.
  • Interaction with CADM1 promotes natural killer (NK) cell cytotoxicity and interferon-gamma secretion by CD8+ T-cells in vitro, and supports NK cell-mediated tumor rejection in vivo. Related references: PMID:15811952
  • CD355/CRTAM regulates CD8+ T-cell proliferation in response to T-cell receptor (TCR) activation but appears dispensable for CD8+ T-cell-mediated cytotoxicity.
  • Binding to SCRIB promotes late-phase polarization of CD4+ T-cells, which in turn modulates TCR-mediated proliferation and production of IL-17 and IL-22.
  • Through interaction with CADM1 on CD8+ dendritic cells, it controls the retention of activated CD8+ T-cells within the draining lymph node.
  • CD355/CRTAM is required for the intestinal retention of intraepithelial CD4+ CD8+ T-cells and contributes to the retention of CD8+ and CD4+ T-cells in the gut, partly by promoting adhesion to CD103+ dendritic cells.
  • Subcellularly, this transmembrane glycoprotein localizes to the cell membrane and is predominantly expressed in activated class-I MHC-restricted T-cells, NKT cells, and CD8+ T-cells, with high abundance in spleen, thymus, and small intestine.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 18-287) corresponds to the predicted extracellular domain of CD355/CRTAM, supporting its use for cell surface detection by flow cytometry.
  • As an -conjugated antibody, no secondary detection reagent is needed; titrate the conjugate to determine the optimal concentration for staining.
  • For positive controls, consider using activated CD8+ T-cells or NK cell lines known to upregulate CD355/CRTAM.
  • The product datasheet indicates a predicted protein weight of 22/44 kDa, reflecting possible isoforms; if adapting for Western blot, validate specificity in the intended sample type.
