Biological Background: CD352/SLAMF6 Function and Localization
- SLAM family member 6 (SLAMF6), also known as NTB-A, is a self-ligand receptor of the signaling lymphocytic activation molecule family. It modulates activation and differentiation of immune cells, bridging innate and adaptive immunity. Related references: PMID:11489943, PMID:16920955, PMID:22184727
- In natural killer (NK) cells, SLAMF6 triggers cytolytic activity when high surface densities of natural cytotoxicity receptors are present, implicating VAV1 phosphorylation; this activation depends on the adapter SH2D1B/EAT-2 via ITSMs. Related references: PMID:16920955
- In T-cells, SLAMF6 promotes differentiation into a Th17 phenotype, increasing IL-17 secretion through recruitment of RORC to the IL-17 promoter; this costimulatory activity requires the adapter SH2D1A/SAP. Related references: PMID:22184727, PMID:22989874
- SLAMF6 is implicated in thymic NKT cell development, controlling the transition from positive selection to expansion/differentiation, possibly in conjunction with SLAMF1.
