Biological Background: CD31/PECAM1 Function and Localization

  • CD31 (PECAM1) is an immunoglobulin superfamily cell adhesion molecule essential for leukocyte transendothelial migration (TEM) under most inflammatory conditions. Phosphorylation of Tyr-690 is critical for TEM and trafficking of the lateral border recycling compartment (LBRC). Related references: PMID:17580308, PMID:19342684
  • Through homophilic interactions, it contributes to endothelial cell–cell adhesion at cell junctions. Heterophilic binding with CD177 facilitates neutrophil transendothelial migration. Related references: PMID:27958302, PMID:17580308
  • Homophilic ligation of PECAM1 prevents macrophage phagocytosis of viable leukocytes by transmitting a detachment signal, while promoting phagocytosis of apoptotic leukocytes by tethering them to phagocytes. Related references: PMID:12110892
  • It modulates bradykinin receptor BDKRB2 activation and regulates bradykinin- and hyperosmotic shock-induced ERK1/2 signaling in endothelial cells. Related references: PMID:18672896
  • CD31 induces susceptibility to atherosclerosis, though it does not directly protect against apoptosis.
  • The protein is predominantly expressed on platelets, leukocytes, and endothelial cell borders. Isoform Long is the major form; Delta12, Delta14–15, Delta14, and Delta15 show tissue-specific distributions (e.g., Delta15 in brain, testis, ovary, and HUVECs).
  • It localizes to the cell membrane, membrane rafts, and cell junctions, consistent with its role in adhesion and signaling.
  • Post-translational modifications include palmitoylation, phosphorylation, and N‑linked glycosylation. The protein contains a signal peptide, multiple immunoglobulin‑like domains, a transmembrane segment, and a cytoplasmic tail with ITIM motifs.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 28–601, corresponding to the extracellular domain; the antibody likely recognizes surface CD31, making it suitable for flow cytometry on live cells.
  • Titrate the conjugate using relevant positive controls (e.g., HUVECs, platelets, leukocyte subsets) and include an appropriate isotype control.
  • For multicolor flow panels, carefully compensate for the signal and avoid fixation protocols that may alter epitope accessibility.
  • Validate the antibody in the specific sample matrix (e.g., whole blood, PBMCs) and consider enzymatic or non‑enzymatic cell harvesting methods that preserve surface CD31.
