Biological Background: CD303/BDCA-2 Function and Localization
- CD303, also known as BDCA-2, dendritic lectin, and CLECSF7, is a member of the C-type lectin family encoded by the CLEC4C gene.
- Serves as a lectin-type cell surface receptor that specifically recognizes non-sialylated galactose-terminated biantennary glycans, binds to serum IgG, and facilitates antigen uptake for peptide presentation to T-cells. Related references: PMID:11748283, PMID:21880719, PMID:25995448
- Plays a role in the inhibition of type I interferon (IFN-α/β) production in plasmacytoid dendritic cells, thereby modulating antiviral innate immunity. Related references: PMID:11748283, PMID:21880719
- Localizes to the cell membrane as a single-pass type II transmembrane protein, anchored via a signal-anchor domain.
- Constitutively expressed in immature monocyte-derived dendritic cells (iMDDC) and significantly down-regulated during LPS-induced maturation, but not by TNF stimulation.
- Contains predicted disulfide bonds and glycosylation sites, contributing to structural stability and lectin domain function.
- May act as a signaling receptor that activates protein-tyrosine kinases and mobilizes intracellular calcium upon ligand engagement. Related references: PMID:11748283
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal staining concentration for your cell type and instrument.
- Since CD303/BDCA-2 is a cell surface antigen, standard live-cell staining protocols without permeabilization are appropriate; verify if fixation affects epitope recognition.
- Include appropriate isotype controls and, when possible, use a peptide blocking experiment to confirm antigen-specific binding.
- Cross-reactivity is documented for human; if testing other species, empirical validation is recommended.
