Biological Background: CD300F/IREM-1 Function and Localization
- Also known as CMRF35-like molecule 1, CD300 antigen-like family member F, IREM-1, IgSF13, NKIR, and CLM1. It belongs to the CD300 family of immunoregulatory receptors, which play diverse roles in immune cell signaling.
- Acts as an inhibitory receptor on myeloid cells and mast cells, delivering negative regulatory signals to control cellular activation. Related references: PMID:15549731
- Recognizes and binds phosphatidylserine (PS) exposed on the surface of apoptotic cells, promoting macrophage-mediated efferocytosis, a process critical for immune homeostasis and resolution of inflammation.
- Negatively regulates Fc epsilon receptor-dependent mast cell activation and allergic responses through interaction with ceramide and sphingomyelin ligands. Related references: PMID:24035150
- Negatively regulates Toll-like receptor (TLR) signaling via activation of the phosphatases SHP-1 and SHP-2, dampening MYD88 and TRIF-dependent pathways. Related references: PMID:22043923
- Expressed as a cell membrane glycoprotein; predicted molecular weight of the core protein is ~32 kDa, with observed isoforms at 18 kDa, 21 kDa, 26 kDa, and 32 kDa due to alternative splicing and glycosylation.
- Highly expressed in spleen, peripheral blood leukocytes, monocytes, and lung; weakly in thymus, heart, brain, placenta, liver, skeletal muscle, kidney, pancreas, prostate, testis, ovary, small intestine, and colon. Expression is selective for monocytes and monocyte-related cells.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 20–156), and the antibody has been tested in flow cytometry (FC). Consider using appropriate isotype controls and validating staining in human monocyte or myeloid cell lines.
- As a rabbit monoclonal antibody, it may offer high specificity and minimal lot-to-lot variation, supporting reproducible FC assays.
- Titrate the antibody to determine the optimal concentration for your specific experimental setup.
- CD300F/IREM-1 is expressed on the surface of myeloid cells and mast cells; ensure proper cell handling and staining protocols to maintain surface antigen integrity.
