Biological Background: CD300a Function and Localization

  • CD300a, also known as CMRF35-like molecule 8 (CLM-8), IRp60, or IgSF12, is an inhibitory receptor belonging to the CD300 family of immunoglobulin superfamily members.
  • It functions as a negative regulator of natural killer (NK) cell cytolytic activity and mast cell degranulation, contributing to immune homeostasis. Related references: PMID:10746781, PMID:16339535, PMID:9701027
  • CD300a negatively regulates Toll-like receptor (TLR) signaling via the MYD88 pathway by activating the phosphatase PTPN6, but does not affect the TRIF pathway. Related references: PMID:22043923
  • The protein is expressed on a broad range of immune cells, including NK cells, T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes, and monocytes.
  • It is localized to the cell membrane as a single-pass transmembrane receptor, with an extracellular immunoglobulin domain and intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that mediate inhibitory signaling.
  • Alternative splicing generates multiple isoforms with predicted molecular weights of approximately 33 kDa, 20 kDa, 12 kDa, and 16.5 kDa (UniProt: 33.2 kDa for canonical form).
  • Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation, which are critical for its structural integrity and function.

Experimental Guidance and Technical Tips

  • The conjugate allows direct detection of CD300a by flow cytometry without a secondary antibody, simplifying the staining protocol and reducing background.
  • As the immunogen corresponds to the extracellular domain (amino acids 18–178), the antibody is expected to bind to the native cell-surface receptor on live cells; consider using appropriate isotype controls and viability dyes in your panel design.
  • For optimal performance, titrate the antibody on your cell type of interest and validate in relevant human immune cell populations such as NK cells, T cells, or mast cells.
  • Avoid fixation with methanol or harsh permeabilization buffers that may alter the epitope; if intracellular staining is required, test with mild permeabilization and confirm retention of signal.
