Biological Background: CD3 epsilon Function and Localization
- CD3 epsilon is the epsilon chain of the T-cell surface glycoprotein CD3, also known as T-cell surface antigen T3/Leu-4 epsilon chain, encoded by the CD3E gene (also designated T3E) in humans.
- As a component of the TCR-CD3 complex on T-lymphocytes, CD3 epsilon plays an essential role in adaptive immune responses by relaying activation signals from the T-cell receptor into the cell. Related references: PMID:15294938, PMID:15546002, PMID:2470098
- The cytoplasmic domain contains immunoreceptor tyrosine-based activation motifs (ITAMs) that become phosphorylated by Src family kinases LCK and FYN upon TCR engagement, initiating downstream signaling cascades. Related references: PMID:2470098, PMID:40592325
- CD3 epsilon also participates in internalization and down-regulation of surface TCR-CD3 complexes via endocytosis sequences in its cytosolic region. Related references: PMID:10384095, PMID:26507128
- Beyond TCR signaling, CD3 epsilon serves as a receptor for ITPRIPL1, which inhibits T-cell activation by promoting NCK1 interaction and blocking downstream ERK-NFkB signaling and calcium influx. Related references: PMID:12110186, PMID:38614099
- The protein localizes to the cell membrane and contains structural features including a signal peptide, an immunoglobulin-like domain, a transmembrane helix, and conserved disulfide bonds.
- The UniProt record lists molecular weight ~23.1 kDa; post-translational modifications include phosphorylation at ITAM tyrosines, which is critical for signal transduction.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–126 of human CD3 epsilon (extracellular domain). For flow cytometry applications, consider a standard cell surface staining protocol using live, unfixed cells to preserve epitope conformation.
- Validate the antibody in a relevant sample system such as human peripheral blood mononuclear cells (PBMCs) or CD3-positive T-cell lines (e.g., Jurkat), ensuring proper isotype control and Fc blocking if necessary.
- Although the antibody is specified for human reactivity, cross-reactivity with other species is not reported; if off-target studies are required, perform sequence alignment and pilot testing.
- Since the product is a rabbit monoclonal IgG, use appropriate anti-rabbit secondary detection reagents when indirect labeling is employed.
