Biological Background: Human CD24 Function and Localization
- CD24 is a heavily glycosylated GPI-anchored cell surface glycoprotein, also known as small cell lung carcinoma cluster 4 antigen. The core protein has a predicted molecular weight of ~8 kDa, but extensive glycosylation produces mature forms of ~13 kDa or larger.
- It may play a pivotal role in cell differentiation across different cell types. Signaling is thought to be triggered by lectin-like ligand binding to CD24 carbohydrates, transduced via GPI-anchor-derived second messengers.
- CD24 modulates B-cell activation responses, promoting antigen-dependent proliferation of B-cells and preventing their terminal differentiation into antibody-forming cells. Related references: PMID:11313396
- In association with SIGLEC10, CD24 may selectively suppress immune responses to danger-associated molecular patterns (DAMPs) such as HMGB1, HSP70, and HSP90, playing a role in the control of autoimmunity (By similarity).
- The protein is localized to the cell membrane via its GPI anchor.
- CD24 is expressed primarily on B-cells, including B-cell lines, erythroleukemia cells, and small cell lung carcinoma cell lines. It is also found on the surface of T-cells.
- Annotated keywords include alternative splicing, glycoprotein, GPI-anchor, lipoprotein, membrane, signal, and reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal extracellular region (aa 1–80), likely targeting an epitope exposed on live cells, making it suitable for surface staining without permeabilization in flow cytometry and immunofluorescence.
- As a directly conjugated product, this antibody eliminates the need for secondary detection steps, reducing background and simplifying multiplex assays.
- Titrate the antibody to determine the optimal working dilution for your specific cell type and experimental conditions.
- Include appropriate positive (e.g., B-cell lines like P32/ISH) and negative (CD24−) controls to validate staining specificity.
- For intracellular detection, permeabilization may be required; consider fixation and permeabilization protocols compatible with the fluorophore.
