Biological Background: BCAM Function and Localization

  • Basal cell adhesion molecule (BCAM), also known as Lutheran blood group glycoprotein or CD239, is a transmembrane glycoprotein that functions as both a receptor and an adhesion molecule, playing a crucial role in cell adhesion, motility, migration, and invasion. Related references: PMID:9616226, PMID:31413112
  • Through its extracellular domain, BCAM binds to extracellular matrix proteins, including laminin alpha-5 (LAMA5), to mediate cell adhesion, while its intracellular domain interacts with cytoskeletal proteins such as hemoglobin to facilitate signal transduction. Related references: PMID:15975931, PMID:17158232
  • JAK2 induces BCAM phosphorylation, activating adhesion to laminin via a Rap1/AKT signaling pathway independently of EPOR. Related references: PMID:23160466
  • BCAM is located at the cell membrane and shows wide tissue distribution, with highest expression in the pancreas and very low levels in the brain. It is closely associated with the basal layer of epithelial cells and the endothelium of blood vessel walls.
  • Post-translational modifications include phosphorylation and disulfide bond formation. BCAM contains immunoglobulin-like domains and carries the Lutheran blood group antigen.

Experimental Guidance and Technical Tips

  • This antibody is conjugated to ; consider titrating for optimal staining index in flow cytometry assays using appropriate human cell lines known to express BCAM.
  • Because BCAM is widely expressed in epithelia and endothelium, include relevant positive and negative controls to confirm specificity.
  • The immunogen corresponds to the extracellular region (amino acids 32-547), supporting recognition of the native cell surface protein.
