Biological Background: CD225/IFITM1 Function and Localization
- IFITM1 (Interferon-induced transmembrane protein 1, also known as CD225, 9-27, IFI17, LEU13, and DSPA2a) is a member of the IFITM family of antiviral restriction factors.
- Exhibits broad antiviral activity by inhibiting viral fusion and release of viral contents into the cytosol; restricts influenza A virus, SARS coronaviruses (SARS-CoV and SARS-CoV-2), filoviruses (Ebola and Marburg), flaviviruses (Dengue and West Nile), HIV-1, and hepatitis C virus (HCV). - Specifically blocks SARS-CoV-2 spike protein-mediated syncytia formation. Related references: PMID:33051876
- Localizes to the cell membrane and lysosome membrane, consistent with its role in trapping endocytosed virions for lysosomal degradation.
- Highly expressed in bone at the protein level; expression is elevated in colon, cervical, esophageal, and ovarian cancers, as well as in glioma cell lines.
- Involved in cell adhesion, growth control, and osteoblast differentiation; mediates IFN-gamma antiproliferative effects via ERK inhibition or p53-dependent G1 arrest. Undergoes palmitoylation and phosphorylation, which likely regulate its membrane association and antiviral function.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-80) of human IFITM1; confirm epitope specificity in your experimental system.
- For immunofluorescence/immunocytochemistry (IF/ICC), include a permeabilization step to detect intracellular IFITM1; cell surface staining may be observed under certain fixation conditions.
- For intracellular flow cytometry (FC intra), optimize fixation and permeabilization protocols; validate staining in relevant human cell lines or primary cells.
- IFITM1 is interferon-inducible; consider pre-treating cells with IFN-alpha or IFN-gamma to upregulate target expression in your assays.
