Biological Background: CD225/IFITM1 Function and Localization

  • IFITM1 (Interferon-induced transmembrane protein 1, also known as CD225, 9-27, IFI17, LEU13, and DSPA2a) is a member of the IFITM family of antiviral restriction factors.
  • Exhibits broad antiviral activity by inhibiting viral fusion and release of viral contents into the cytosol; restricts influenza A virus, SARS coronaviruses (SARS-CoV and SARS-CoV-2), filoviruses (Ebola and Marburg), flaviviruses (Dengue and West Nile), HIV-1, and hepatitis C virus (HCV).   • Specifically blocks SARS-CoV-2 spike protein-mediated syncytia formation. Related references: PMID:33051876
  • Localizes to the cell membrane and lysosome membrane, consistent with its role in trapping endocytosed virions for lysosomal degradation.
  • Highly expressed in bone at the protein level; expression is elevated in colon, cervical, esophageal, and ovarian cancers, as well as in glioma cell lines.
  • Involved in cell adhesion, growth control, and osteoblast differentiation; mediates IFN-gamma antiproliferative effects via ERK inhibition or p53-dependent G1 arrest. Undergoes palmitoylation and phosphorylation, which likely regulate its membrane association and antiviral function.
