Biological Background: IFITM1 Function and Localization
- IFITM1 is an interferon-induced transmembrane protein that belongs to the dispanin family (subfamily A member 2a) and is also known as CD225, Leu-13, or 9-27.
- Functions as a broad-spectrum antiviral restriction factor by blocking viral entry at the endosomal/lysosomal stage; it permits endocytosis but prevents viral fusion, thereby inhibiting viruses such as influenza A, SARS-CoV-1, SARS-CoV-2, Ebola, Marburg, HIV-1, HCV, Dengue, and West Nile virus. - Inhibits SARS-CoV-2 spike-mediated syncytia formation, contributing to restriction of cell-cell fusion. Related references: PMID:33051876

- Localizes to the cell membrane and lysosome membrane, consistent with its role in viral entry restriction.
