Biological Background: IFITM1 Function and Localization

  • IFITM1 is an interferon-induced transmembrane protein that belongs to the dispanin family (subfamily A member 2a) and is also known as CD225, Leu-13, or 9-27.
  • Functions as a broad-spectrum antiviral restriction factor by blocking viral entry at the endosomal/lysosomal stage; it permits endocytosis but prevents viral fusion, thereby inhibiting viruses such as influenza A, SARS-CoV-1, SARS-CoV-2, Ebola, Marburg, HIV-1, HCV, Dengue, and West Nile virus. Related references: PMID:26354436, PMID:33270927
  • Inhibits SARS-CoV-2 spike-mediated syncytia formation, contributing to restriction of cell-cell fusion. Related references: PMID:33051876
  • In hepatocytes, IFITM1 coordinates with IFITM2/3 to target HCV virions for lysosomal degradation. Related references: PMID:26354436
  • Localizes to the cell membrane and lysosome membrane, consistent with its role in viral entry restriction.
  • Additional roles: IFN-gamma-induced antiproliferative effects via ERK inhibition or p53-dependent G1 arrest; positive regulation of osteoblast differentiation; and clinical relevance in cancer, with elevated expression in colon, cervical, esophageal, ovarian cancers, and gliomas.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal 1-80 amino acids, which includes the extracellular domain; consider this when designing blocking peptides or epitope mapping experiments.
  • Since this antibody is validated for intracellular flow cytometry, ensure appropriate permeabilization when detecting IFITM1, as the protein localizes to the cell membrane and endosomal/lysosomal compartments.
  • IFITM1 expression is interferon-inducible; consider stimulating cells with IFN-α/γ to upregulate the target in your experimental system.
  • Given its role in viral entry, this antibody may be useful for studying viral restriction mechanisms in cell culture models.
