Biological Background: CD177 Function and Localization

  • CD177, also known as HNA-2a, NB1 GP, or PRV-1, is a glycosylphosphatidylinositol (GPI)-anchored glycoprotein expressed on the surface of a subset of human neutrophils.
  • It acts as a receptor for proteinase 3 (PRTN3), displaying the protease on the neutrophil surface and potentially enhancing endothelial junctional integrity during neutrophil diapedesis. Related references: PMID:17244676 PMID:18462208 PMID:23202369
  • In association with beta-2 integrins ITGAM/CD11b and ITGB2/CD18, CD177 mediates TNF-primed neutrophil activation, including degranulation and superoxide production, while attenuating chemokine signaling to favor adhesion over migration. Related references: PMID:21193407 PMID:28807980
  • Heterophilic interaction with PECAM1 on endothelial cells contributes to neutrophil transendothelial migration in vitro, though in vivo studies suggest a dispensable role in bacterial infection-induced recruitment. Related references: PMID:17580308 PMID:23461681
  • Highly expressed in normal bone marrow, CD177 expression begins at the metamyelocyte stage and is restricted to a subset of mature neutrophils, with inter-individual variability in the percentage of positive cells. Related references: PMID:10753836 PMID:12377969 PMID:17244676 PMID:18462208 PMID:24926686
  • Subcellularly, CD177 is found on the cell membrane, in membrane rafts, secretory granules, and lamellipodia of neutrophils.
  • Alternative splicing and extensive glycosylation yield isoforms with apparent molecular weights ranging from 15 to 46 kDa, as observed in western blotting.
  • As a member of the immunoglobulin superfamily with a GPI anchor, CD177 is implicated in innate immunity and cell adhesion processes.

Experimental Guidance and Technical Tips

  • The immunogen comprises a recombinant fusion protein covering the extracellular domain (amino acids 22-407), targeting the mature, surface-exposed region of CD177.
  • For flow cytometry, consider titrating the antibody to determine the optimal concentration for your specific cell type and experimental setup.
  • Validate the antibody in your sample system using appropriate controls, such as isotype controls and known CD177-expressing neutrophil subsets.
  • The fluorophore is compatible with standard red laser (561–594 nm) configurations; ensure your cytometer is equipped with the appropriate excitation and emission filters.
