Biological Background: CD172a/SIRPα Function and Localization

  • CD172a/SIRPα (also known as BIT, MFR, P84, SIRP, MYD-1, SHPS1, PTPNS1) is an immunoglobulin-like cell surface receptor for CD47, encoded by the SIRPA gene. It acts as a docking protein, inducing translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane.
  • It supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment, and may play a key role in intracellular signaling during synaptogenesis and in synaptic function (By similarity).
  • SIRPα is involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin, and mediates negative regulation of phagocytosis, mast cell activation and dendritic cell activation.
  • CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells. SIRPα also plays a role in antiviral immunity by limiting new world arenavirus infection through decreased virus internalization (By similarity).
  • SIRPα is a receptor for THBS1. Interaction with THBS1 stimulates phosphorylation of SIRPα and is involved in ROS signaling in non-phagocytic cells, stimulating NADPH oxidase-derived ROS production. Related references: PMID:24511121
  • The protein is a single-pass type I membrane protein and is ubiquitously expressed, with high expression in brain. It is detected on myeloid cells but not T-cells, and at lower levels in heart, placenta, lung, testis, ovary, colon, liver, small intestine, prostate, spleen, kidney, skeletal muscle and pancreas.
  • Key structural features include alternative splicing, disulfide bonds, glycosylation, immunoglobulin domains, and a transmembrane helix. It is also a phosphoprotein and contains an SH3-binding domain.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein covering the extracellular region (amino acids 31-370) of human SIRPα, which contains the ligand-binding immunoglobulin domains. This design should generate antibodies that recognize native, non-denatured epitopes, making this clone suitable for FC applications.
  • For flow cytometry, consider titrating the antibody to determine the optimal working concentration for your specific cell type. Since SIRPα is highly expressed on myeloid cells, it can serve as a lineage marker in human immune cell profiling.
  • Although the antibody is validated for human cross-reactivity, cross-reactivity with other species has not been tested. Validate in the relevant sample system if cross-species application is needed.
