Biological Background: CD166/ALCAM Function and Localization

  • CD166/ALCAM is a cell adhesion molecule of the immunoglobulin superfamily, also known as activated leukocyte cell adhesion molecule (ALCAM) and MEMD, encoded by the ALCAM gene on human chromosome 3.
  • It mediates both heterotypic (with CD6) and homotypic cell-cell contacts, promoting T-cell activation and proliferation, and contributing to the formation and maturation of the immunological synapse. Related references: PMID:15048703 PMID:16352806 PMID:24945728 PMID:15294938
  • Through homotypic interactions, ALCAM inhibits endothelial cell migration and promotes tube formation, and mediates dendritic cell attachment to endothelial cells. Related references: PMID:15496415 PMID:23169771
  • It is required for normal hematopoietic stem cell engraftment in the bone marrow and for maintaining normal numbers of hematopoietic stem cells. Related references: PMID:24740813
  • Acts as a ligand for the LILRB4 receptor, thereby enhancing LILRB4-mediated inhibition of T cell proliferation. Related references: PMID:29263213
  • An alternative splicing isoform functions as a decoy receptor, competing for the same interaction partners and inhibiting the activities of the membrane-bound isoforms; it promotes endothelial cell migration and inhibits tube formation.
  • The protein localizes to the cell membrane and cell projections (axons and dendrites); a secreted isoform has also been identified.
  • Detected in hematopoietic stem cells, lymph vessel endothelial cells, activated T and B cells, monocytes, thymic epithelial cells, neurons in the brain, and certain tumor cell lines (e.g., highly metastasizing melanoma). Related references: PMID:24740813 PMID:23169771 PMID:9502422 PMID:7760007

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 28–526) of human CD166/ALCAM, making this conjugate suitable for detecting surface expression by flow cytometry.
  • As a directly conjugated primary antibody (), no secondary reagent is required; protect from light during handling and staining to maintain fluorophore performance.
  • For optimal flow cytometry results, titrate the antibody to determine the best concentration for your specific cell type and experimental conditions.
  • Include appropriate isotype controls and fluorescence minus one (FMO) controls to distinguish specific binding.
  • While validated for FC, the antibody may also be suitable for other applications like immunofluorescence or IHC, provided thorough validation in the relevant sample system is performed.
