Biological Background: CD15/SSEA-1 (FUT4) Function and Localization

  • The protein encoded by FUT4 (gene ID 2526) is Alpha-(1,3)-fucosyltransferase 4, a glycosyltransferase responsible for the final fucosylation step that creates the Lewis X (CD15/SSEA-1) carbohydrate antigen on N‑ and O‑linked glycoproteins. Related references: PMID:1702034 PMID:1716630 PMID:29593094

  • Future 4 transfers fucose from GDP‑β‑L‑fucose to the distal N‑acetylglucosamine of type‑2 lactosamine chains (Galβ1‑4GlcNAc), most efficiently on non‑sialylated acceptors, thereby generating the Lewis X epitope. Related references: PMID:1716630 PMID:29593094

  • In certain cell types, FUT4 can also fucosylate an internal GlcNAc residue to produce the VIM‑2 antigen, a ligand for E‑selectin (SELE). Related references: PMID:11278338 PMID:1716630

  • Together with FUT7, FUT4 contributes to the biosynthesis of selectin ligands (sialyl Lewis X and 6‑sulfo sialyl Lewis X) that mediate leukocyte rolling and homing to sites of inflammation.

  • FUT4 is a transmembrane protein that resides in the Golgi stack membrane, where it encounters its glycoprotein substrates during transit through the secretory pathway.

  • Expression has been detected in bone marrow‑derived mesenchymal stem cells, cord blood immature promyelocytes, and in peripheral blood myeloid and lymphoid populations.

  • The protein contains alternative initiation sites and is itself N‑glycosylated; it belongs to the glycosyltransferase 10 family and contains a signal‑anchor sequence.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide derived from human CD15/SSEA-1; consider that the antibody may recognize a linear epitope that mimics the native carbohydrate structure or a peptide backbone of the carrier glycoprotein.
  • For flow cytometry, include appropriate isotype controls and verify reactivity on a CD15‑positive cell line (e.g., HL‑60) and a CD15‑negative line (e.g., K562).
  • For immunophenotyping applications, titrate the antibody to determine the optimal concentration for your specific cell population and staining protocol.
  • The predicted molecular weight of the target enzyme is ~59 kDa; in western blotting (if attempted) be aware that the antibody may detect either the glycosylated FUT4 protein or its glycoprotein products bearing CD15.
