Biological Background: Syndecan-1 Function and Localization

  • Syndecan-1 (also known as CD138, SDC1, SYND1) is a transmembrane heparan sulfate proteoglycan that belongs to the syndecan family.
  • It links the cytoskeleton to the interstitial matrix through its heparan sulfate and chondroitin sulfate glycosaminoglycan chains (By similarity).
  • Syndecan-1 regulates exosome biogenesis in concert with SDCBP and PDCD6IP. Related references: PMID:22660413
  • It can induce its own expression in dental mesenchymal and neighboring epithelial cells via an MSX1-mediated pathway (By similarity).
  • Subcellularly, Syndecan-1 localizes to the plasma membrane, is secreted, and is found in extracellular exosomes.
  • Tissue expression has been detected in placenta and in fibroblasts at the protein level. Related references: PMID:32337544, PMID:36213313
  • Post-translational modifications include glycosylation (heparan sulfate and chondroitin sulfate attachment), phosphorylation, and proteolytic shedding.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 23–251) of human CD138/Syndecan-1, suggesting the antibody recognizes the exposed ectodomain on the cell surface.
  • Directly conjugated to , this antibody is validated for flow cytometry (FC); consider optimizing antibody titration and including appropriate isotype and FMO controls.
  • The core protein mass is approximately 32 kDa, but extensive glycosylation often results in a higher apparent molecular weight in denaturing gels.
  • Antigen retrieval or fixation protocols should be tested to ensure epitope preservation, as harsh conditions may alter the extracellular domain conformation.
