Biological Background: Syndecan-1 Function and Localization
- Syndecan-1 is a cell surface proteoglycan that carries both heparan sulfate and chondroitin sulfate chains, serving as a critical link between the cytoskeleton and the interstitial matrix.
- In collaboration with SDCBP and PDCD6IP, syndecan-1 plays a key role in the regulation of exosome biogenesis. Related references: PMID:22660413
- Syndecan-1 can induce its own expression in dental mesenchymal cells and neighboring dental epithelial cells through an MSX1-mediated pathway.
- The protein is localized to the membrane and is also secreted, notably into extracellular exosomes.
- Protein-level expression of syndecan-1 has been detected in placenta and fibroblasts.
- Syndecan-1 undergoes post-translational modifications including phosphorylation and glycosylation, and is classified as a glycoprotein, proteoglycan, and phosphoprotein.
Experimental Guidance and Technical Tips
- This antibody is generated against a recombinant fusion protein encompassing the extracellular domain of human CD138 (amino acids 23-251), ensuring specificity for the native protein.
- The PE conjugation allows direct detection in flow cytometry without the need for a secondary antibody; consider titrating the antibody to optimize signal-to-noise ratio.
- Validate the antibody in the relevant human sample system, as cross-reactivity with other species has not been reported.
