Biological Background: Semaphorin-4D Function and Localization
- Semaphorin-4D (SEMA4D), also known as CD100, A8, BB18, or GR3, is a member of the semaphorin family encoded by the SEMA4D gene. Alternative splicing produces isoforms; the canonical isoform has a molecular weight of ~96 kDa.
- Functions as a receptor for plexins PLXNB1 and PLXNB2, playing a critical role in cell-cell signaling. Related references: PMID:20877282
- In the nervous system, SEMA4D regulates GABAergic synapse development and modulates the complexity and arborization of developing neurites in hippocampal neurons via PLXNB1-mediated RHOA activation. Related references: PMID:19788569
- Promotes the migration of cerebellar granule cells and induces endothelial cell migration through activation of PTK2B/PYK2, SRC, and the phosphatidylinositol 3-kinase-AKT pathway. Related references: PMID:16055703
- In the immune system, SEMA4D induces B-cell aggregation and enhances their viability in vitro. Related references: PMID:8876214
- Localizes to the cell membrane and is also secreted into the extracellular space.
- Strongly expressed in skeletal muscle, peripheral blood lymphocytes, spleen, and thymus; lower expression in testes, brain, kidney, small intestine, prostate, heart, placenta, lung, and pancreas. Not detected in colon and liver.
- Contains an immunoglobulin domain, is glycosylated, phosphorylated, and has a disulfide bond. Involved in neurogenesis, differentiation, and developmental processes.
Experimental Guidance and Technical Tips
- The immunogen encompasses the entire extracellular region (aa 22-734) of human SEMA4D, suggesting the antibody likely recognizes an epitope in the extracellular domain. For immunofluorescence, consider using appropriate permeabilization to detect membrane-bound protein; mild permeabilization may also reveal intracellular pools if present.
- For flow cytometry with the conjugate, titrate the antibody to determine the optimal working concentration for your cell type. is bright and photostable, suitable for multiplex panels.
- Since SEMA4D is expressed on immune cells and certain tissues, validate the antibody in the relevant sample system, particularly for tissue-specific expression patterns.
- Note that SEMA4D can be secreted; differentiate surface and intracellular staining by optimizing fixation/permeabilization protocols when needed.
