Biological Background: B7-H2/CD275 (ICOSL) Function and Localization

  • ICOS ligand (ICOSL, also known as B7-H2, CD275, B7RP-1, GL50) is a member of the B7 family of immune regulatory proteins, providing costimulatory signals essential for T-cell activation and effector functions.
  • The interaction between ICOSL and its receptor ICOS on T-cells promotes T-cell proliferation and cytokine secretion, such as interleukin-4 (IL-4) and interferon-gamma (IFN-γ). Related references: PMID:11007762 PMID:11023515 PMID:30498080
  • ICOSL also induces B-cell proliferation and differentiation into plasma cells, highlighting its role in humoral immunity.
  • In endothelial cells, ICOSL is required for proper neutrophil transmigration in response to chemoattractants like CXCL8/IL8 or fMLP, linking adaptive and innate immunity. Related references: PMID:30498080
  • The protein is expressed on the cell membrane and is widely distributed in tissues including lymph nodes, leukocytes, spleen, and on activated monocytes and dendritic cells, as well as peripheral blood B-cells and monocytes.
