Biological Background: B7-H2/CD275 (ICOSL) Function and Localization
- ICOS ligand (ICOSL, also known as B7-H2, CD275, B7RP-1, GL50) is a member of the B7 family of immune regulatory proteins, providing costimulatory signals essential for T-cell activation and effector functions.
- The interaction between ICOSL and its receptor ICOS on T-cells promotes T-cell proliferation and cytokine secretion, such as interleukin-4 (IL-4) and interferon-gamma (IFN-γ). Related references: PMID:11007762 PMID:11023515 PMID:30498080
- ICOSL also induces B-cell proliferation and differentiation into plasma cells, highlighting its role in humoral immunity.
- In endothelial cells, ICOSL is required for proper neutrophil transmigration in response to chemoattractants like CXCL8/IL8 or fMLP, linking adaptive and innate immunity. Related references: PMID:30498080
- The protein is expressed on the cell membrane and is widely distributed in tissues including lymph nodes, leukocytes, spleen, and on activated monocytes and dendritic cells, as well as peripheral blood B-cells and monocytes.
- ICOSLG undergoes alternative splicing, producing isoforms, and contains immunoglobulin-like domains, a signal peptide, and a transmembrane helix. It is a glycoprotein with disulfide bonds.
Experimental Guidance and Technical Tips
- For flow cytometry, use the directly conjugated antibody for surface staining of live cells without permeabilization, as the immunogen targets the extracellular domain (aa 19-256).
- Titrate the antibody to determine the optimal concentration for your specific cell type and experimental setup; a typical starting range is 0.1–1 µg per 10^6 cells.
- Include an appropriate isotype control (rabbit IgG ) to assess non-specific binding.
- Validate reactivity in your target cell population expressing B7-H2/CD275, such as activated monocytes, dendritic cells, or B-cell lines, and confirm specificity using ICOSL knockout or blocking controls where possible.
