Biological Background: HTRA2 Function and Localization
- HTRA2 is a mitochondrial serine protease that promotes apoptosis by cleaving and inactivating inhibitor of apoptosis proteins (IAPs) such as BIRC6, enhancing caspase activity (PubMed:10873535, PubMed:15200957, PubMed:36758104, PubMed:36758105). Related references: PMID:10873535 PMID:15200957 PMID:36758104 PMID:36758105
- It also triggers apoptosis via a caspase-independent mechanism that depends on its serine protease activity (PubMed:15200957). Related references: PMID:15200957
- The canonical isoform is catalytically active; however, alternative splicing generates an isoform that appears proteolytically inactive.
- HTRA2 resides primarily in the mitochondrial intermembrane space and can associate with the mitochondrial membrane; it is also detected in the endoplasmic reticulum.
- It is ubiquitously expressed in human tissues.
- Cleaves THAP5 and promotes its degradation during apoptosis (PubMed:19502560). Related references: PMID:19502560
- Keywords: Serine protease, Apoptosis, Neurodegeneration, Parkinson disease, Parkinsonism, Epilepsy, Alternative splicing, Ubl conjugation, Zymogen.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 100-200 of human HTRA2; cross-reactivity with mouse and rat suggests sequence conservation in this region.
- For Western blot, a band of approximately 49 kDa is expected in samples from human, mouse, and rat.
- ELISA applications should be optimized; consider testing with recombinant HTRA2 or cell lysates.
- Due to HTRA2's role in apoptosis, expression levels may vary under different cellular conditions; include appropriate positive and negative controls.
