Biological Background: HSPA2 Function and Localization
- HSPA2 (Heat shock-related 70 kDa protein 2, also known as HSP70-2 or HSP70-3) is a member of the heat shock protein family A (Hsp70) that functions as a molecular chaperone.
- It plays a central role in protein quality control by facilitating proper protein folding, refolding of misfolded proteins, and targeting of damaged proteins for degradation (PubMed:26865365). Related references: PMID:26865365
- Chaperone activity is coupled to an ATPase cycle: ATP-bound HSPA2 has low substrate affinity, and ATP hydrolysis to ADP induces a conformational change that increases substrate binding, enabling cycles of substrate binding and release.
- HSPA2 is essential for spermatogenesis, and in association with SHCBP1L, it may help maintain spindle integrity during meiosis in male germ cells.
- Subcellularly, HSPA2 localizes to the cytoplasm, cytoskeleton, and mitotic/meiotic spindle, consistent with its role in meiosis.
- Post-translational modifications include phosphorylation and methylation, which may regulate its activity and interactions.
- HSPA2 is involved in the cellular stress response, protecting the proteome from stress-induced damage.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 400-639) targets the substrate-binding domain of HSPA2, which is relatively conserved; however, the antibody may also recognize other Hsp70 family members with high homology in this region. Consider sequence alignment to assess potential cross-reactivity if specificity is critical.
- For Western blot, the predicted molecular weight is approximately 70 kDa. Validate in relevant cell lines or tissues known to express HSPA2 (e.g., testis, certain cancer cells) using appropriate loading controls.
- In IF/ICC, anticipate cytoplasmic and spindle staining at specific cell cycle stages (e.g., meiotic cells). Include negative controls (e.g., HSPA2 knockdown/knockout samples) to confirm specificity.
- For ELISA, titrate the antibody to optimize signal-to-noise ratio, and pair with a suitable detection system.
