Biological Background: HSP70 (HSPA1A/HSPA1B) Function and Localization
- Heat shock 70 kDa protein 1A (HSPA1A) and 1B (HSPA1B) are highly homologous molecular chaperones involved in protein folding, trafficking, and degradation.
- Acts as an ATP-dependent chaperone, cycling between low-affinity (ATP-bound) and high-affinity (ADP-bound) states for substrate proteins, regulated by co-chaperones including J-domain proteins, nucleotide exchange factors (BAG family), and TPR domain chaperones like HOPX and STUB1. Related references: PMID:24012426, PMID:24318877, PMID:26865365
- Acetylation/deacetylation switches HSP70 function between protein refolding (acetylated, binds HOPX) and degradation (deacetylated, binds STUB1/CHIP). Related references: PMID:27708256
- Maintains proteostasis during stress; enhances STUB1-mediated ubiquitination of signaling proteins SMAD3 and FOXP3, modulating TGF-beta and Treg pathways. Related references: PMID:24613385, PMID:23973223
- Required for centrosome integrity and bipolar spindle assembly during mitosis. Related references: PMID:27137183
- Negatively regulates HSF1 transcriptional activity to attenuate heat shock response. Related references: PMID:9499401
- Clears misfolded proteins like PRDM1 via SYNV1/HRD1‑mediated proteasomal degradation. Related references: PMID:28842558
- (Microbial infection) Functions as a post-attachment receptor for rotavirus A entry.
- Subcellularly located in cytoplasm, nucleus, centrosome, and can be secreted; post-translationally modified by acetylation, methylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 500-600 of human HSP70 (NP_005336.3), a region conserved across species, supporting cross-reactivity with human, mouse, rat, wheat, and maize.
- Suitable for Western blot (WB), immunohistochemistry (IHC-P), immunofluorescence (IF/ICC), and ELISA. Optimize antibody dilution for each assay and sample type.
- For WB, the expected band size is approximately 70 kDa; include appropriate positive and negative controls.
- Validate in the relevant cell or tissue system, especially under heat shock or stress conditions that upregulate HSP70.
