Biological Background: HSP60/HSPD1 Function and Localization
- HSP60, encoded by the HSPD1 gene, is a 60 kDa mitochondrial chaperonin also known as Chaperonin 60 (CPN60), heat shock protein 60 (HSP-60), and HuCHA60. It belongs to the chaperonin family and functions as a key protein folding catalyst.
- Within the mitochondrial matrix, HSP60 cooperates with its co-chaperonin Hsp10 to facilitate correct folding of newly imported proteins, preventing misfolding and promoting refolding under stress conditions. Related references: PMID:11422376, PMID:1346131
- The functional chaperonin complex comprises two heptameric rings of HSP60 that form a back-to-back double-ring structure. Substrate protein binds one ring, followed by ATP binding and association with two heptameric Hsp10 rings, creating an encapsulated folding chamber.
- Subcellularly, HSP60 is localized exclusively to the mitochondrial matrix, where it ensures proper protein homeostasis and mitochondrial function.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin conjugation, and alternative splicing produces isoforms. Mutations in HSPD1 are linked to neurodegenerative disorders such as hereditary spastic paraplegia and leukodystrophy.

- The immunogen used is a synthetic peptide; therefore, the antibody likely recognizes a linear epitope. Optimize antigen retrieval for IHC-P and fixation conditions for IF/ICC accordingly.
- In Western blot, expect a band at approximately 61 kDa. Use appropriate positive and negative controls (e.g., mitochondrial extracts) and consider siRNA or knockout validation to confirm specificity.
- For IHC-P and IF/ICC, pre-adsorption with the immunogen peptide can help verify signal specificity.
- Cross-reactivity is reported for human, mouse, rat, and wheat, but independent verification in your sample type is recommended.
