Biological Background: Hormone-sensitive lipase Function and Localization
- Hormone-sensitive lipase (HSL), encoded by the LIPE gene, is a multifunctional enzyme with broad substrate specificity, catalyzing the hydrolysis of triacylglycerols (TAGs), diacylglycerols (DAGs), monoacylglycerols (MAGs), cholesteryl esters, and retinyl esters. It shows preferential hydrolysis of DAGs over TAGs and MAGs, and preferentially hydrolyzes the sn-3 position in DAGs and sn-1/sn-2 positions in TAGs.

-
HSL hydrolyzes 2-arachidonoylglycerol, an endocannabinoid, and 2-acetyl monoalkylglycerol ether, the penultimate precursor of platelet-activating factor, linking HSL to endocannabinoid signaling and inflammatory mediator synthesis.
-
In adipose tissue and heart, HSL primarily releases free fatty acids from stored triglycerides for energy; in steroidogenic tissues, it converts cholesteryl esters to free cholesterol for steroid hormone production.
-
Subcellularly, HSL localizes to the cell membrane, caveolae, cytoplasm, cytosol, and the surface of lipid droplets, reflecting its dynamic role in lipid mobilization.
-
Tissue expression is notably high in testis, but HSL is functionally important in multiple metabolic tissues.
-
As a key lipid hydrolase, HSL is implicated in obesity, diabetes mellitus, and cholesterol/steroid metabolism disorders.
Experimental Guidance and Technical Tips
-
The immunogen is a synthetic peptide from the C-terminal region (aa 900-1000), ensuring specific recognition of HSL. For Western blot, expect a band at approximately 117 kDa; validate using adipose tissue or other HSL-expressing cell lysates.
-
For ELISA, consider using the immunizing peptide or full-length recombinant HSL protein to optimize assay conditions and antibody titration.
-
As a rabbit polyclonal antibody, lot-to-lot variability may occur; each new lot should be validated in your specific sample matrix and application.
-
Cross-reactivity with mouse and rat indicates suitability for rodent models; include appropriate species-matched controls.
