Biological Background: HSD3B1 Function and Localization

  • HSD3B1, also known as 3 beta-hydroxysteroid dehydrogenase/Delta 5-->4-isomerase type 1 (3β-HSD I), is encoded by the HSD3B1 gene (aliases: HSDB3A, 3BH, SDR11E1).
  • It is a bifunctional enzyme essential for steroid hormone biosynthesis, catalyzing the oxidation and isomerization of 3β-hydroxy-Δ5-steroid precursors to 3-oxo-Δ4-steroids. Related references: PMID:1401999 PMID:2139411
  • Specifically converts pregnenolone to progesterone, 17α-hydroxypregnenolone to 17α-hydroxyprogesterone, DHEA to androstenedione, and androstenediol to testosterone.
  • Also interconverts 3β-hydroxy and 3-oxo-5α-androstane steroids, controlling active androgen bioavailability; for example, it converts dihydrotestosterone (DHT) to inactive 5α-androstanediol.
  • Subcellular localization includes the endoplasmic reticulum membrane and mitochondrial membrane.
  • Highly expressed in placenta, skin, and mammary gland tissue.
  • Classified under keywords: multifunctional enzyme, steroidogenesis, lipid metabolism, NAD/NADP, and transmembrane.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 250–350 of human HSD3B1; therefore, the antibody likely recognizes an internal epitope and may be sensitive to conformational changes.
  • For Western blotting, a band near 42 kDa is expected. Validate detection in the relevant sample system and consider using appropriate positive controls (e.g., placental or mammary tissue lysates).
  • In IHC-P and ELISA, antigen retrieval and blocking conditions may need optimization. The antibody cross-reacts with mouse and rat, enabling multi-species studies.
