  • HSD3B1 is a bifunctional enzyme belonging to the 3-beta hydroxysteroid dehydrogenase/isomerase family. It catalyzes the oxidation and isomerization of 3β-hydroxy-Δ5-steroid precursors to 3-oxo-Δ4-steroids, an essential step in the biosynthesis of all major steroid hormones. This includes the conversion of pregnenolone to progesterone, 17α-hydroxypregnenolone to 17α-hydroxyprogesterone, dehydroepiandrosterone (DHEA) to 4-androstenedione, and androstenediol to testosterone.   • Beyond de novo steroidogenesis, HSD3B1 regulates local androgen bioavailability by interconverting active and inactive 5α-reduced androgens.
