Biological Background: HSD17B8 Function and Localization
- HSD17B8 (17-beta-hydroxysteroid dehydrogenase 8), also known as KE6, FABG, and SDR30C1, is a 27 kDa member of the short-chain dehydrogenase/reductase (SDR) family.
- Acts as the alpha subunit of the mitochondrial 3-ketoacyl-ACP reductase (KAR) complex, serving as a scaffold protein essential for the solubility and stability of the beta subunit CBR4, thereby enabling mitochondrial fatty acid biosynthesis. Related references: PMID:19571038, PMID:25203508, PMID:30508570
- Catalyzes the NAD-dependent conversion of (3R)-3-hydroxyacyl-CoA to 3-ketoacyl-CoA with no chain length preference; cooperates with enoyl-CoA hydratase 1 as an alternative route for cis-polyunsaturated fatty acid breakdown. Related references: PMID:19571038, PMID:30508570
- Functions as a 17-beta-hydroxysteroid dehydrogenase with highest activity toward estradiol, exhibiting primarily oxidative but conditionally reductive activity to convert estrone to estradiol. Related references: PMID:17978863
- Localized to the mitochondrial matrix.
- Widely expressed, with high abundance in prostate, placenta, and kidney; detected at protein level in brain, cerebellum, heart, lung, ovary, testis, and adrenals. Related references: PMID:17978863, PMID:30508570
- Identified as a phosphoprotein with a known 3D structure; participates in fatty acid and steroid biosynthesis.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering full-length human HSD17B8 (aa 1–261); this design may detect all described isoforms.
- For Western blot, a band at ~27 kDa is expected; verify specificity using mitochondrial lysates from human, mouse, or rat tissues.
- In immunofluorescence/ICC, expect a mitochondrial staining pattern; include appropriate controls and validate in relevant cell systems.
- For ELISA applications, optimize antigen coating and antibody dilution empirically; cross-reactivity with mouse and rat homologs can aid multi-species assays.
