Biological Background: HSD17B7 Function and Localization
- HSD17B7 (also known as PRAP, SDR37C1) is a bifunctional enzyme catalyzing 3‑ketosteroid reduction and 17β‑hydroxysteroid dehydrogenase activity, linking steroid hormone metabolism and cholesterol biosynthesis.
Related references: PMID:11165030, PMID:12574203, PMID:12732193 - It reduces estrone (E1) to the more potent 17β‑estradiol (E2) and inactivates dihydrotestosterone (DHT) to 5α‑androstane‑3β,17β‑diol, modulating estrogen and androgen signaling.
Related references: PMID:12574203, PMID:12732193, PMID:19772289 - The enzyme participates in the post‑squalene cholesterol biosynthesis pathway by reducing zymosterone and other intermediates.
Related references: PMID:11165030, PMID:12829805, PMID:20659585 - HSD17B7 is an integral membrane protein of the endoplasmic reticulum, with a single transmembrane domain.
- It is widely expressed, with highest levels in adrenal gland, liver, lung, and thymus; also found in reproductive tissues, kidney, and the eye ciliary epithelium.
- The protein is subject to alternative splicing and N‑glycosylation, which may regulate its activity or localization.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 110–200) lies within the catalytic domain; as a polyclonal antibody, it may recognize multiple epitopes, including isoforms.
- For Western blot, the predicted molecular weight is 38 kDa; consider that post‑translational modifications (glycosylation) or alternative splicing may shift the observed band.
- In immunohistochemistry (IHC‑P), validate with known positive tissues (e.g., adrenal gland, liver) and include appropriate controls due to potential cross‑reactivity with other short‑chain dehydrogenase/reductase family members.
- For ELISA, the antibody can be used as capture or detection reagent; performance should be titrated in the relevant sample matrix.
