Biological Background: Hsc70-interacting protein Function and Localization
- Hsc70-interacting protein (HIP), also designated ST13, is a cytoplasmic co-chaperone that directly interacts with the molecular chaperone HSC70.
- One HIP oligomer binds the ATPase domains of at least two HSC70 molecules, stabilizing the ADP-bound state with high substrate affinity (By similarity).
- The protein is subject to post-translational modifications including acetylation and phosphorylation, which may regulate its chaperone activity.
- HIP is also known as progesterone receptor-associated p48, aging-associated protein 2, and renal carcinoma antigen NY-REN-33, suggesting roles in diverse cellular pathways.
- According to UniProt, HIP contains TPR repeat domains and has been identified by direct protein sequencing.
- Subcellular localization is predominantly cytoplasmic.
Experimental Guidance and Technical Tips
- Since the immunogen spans the full-length human HIP protein (aa 1-369), the antibody may recognize native and denatured forms; however, for Western blot, we recommend denaturing and reducing conditions with appropriate blocking.
- The reported cross-reactivity with mouse and rat allows use in common model organisms; nonetheless, researchers should validate performance in their specific samples.
