Biological Background: HRAS Function and Localization
- HRAS encodes the GTPase HRas, also known as H-Ras-1, Ha-Ras, transforming protein p21, c-H-ras, and p21ras.
- It functions as a signal transducer in the Ras-MAPK signaling pathway, playing a critical role in the regulation of cell proliferation and survival. Related references: PMID:22821884
- Ras proteins bind GDP/GTP and possess intrinsic GTPase activity, allowing them to act as molecular switches. Related references: PMID:12740440, PMID:14500341, PMID:9020151
- HRas is ubiquitinated by the LZTR1-BCR E3 ubiquitin ligase complex, which regulates its stability and signaling output. Related references: PMID:40934300
- The protein localizes to multiple cellular compartments including the cell membrane, Golgi apparatus membrane, nucleus, cytoplasm, and perinuclear region.
- It is widely expressed across human tissues.
- Post-translational modifications such as palmitoylation, prenylation, acetylation, methylation, and ubiquitination modulate its localization and activity.
- HRAS is a proto-oncogene, and mutations in this gene are associated with various cancers and developmental disorders (RASopathies).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide within the N-terminal region (amino acids 1-100) of human HRAS. Sequence conservation supports expected cross-reactivity with mouse and rat, as indicated.
- For Western blot (WB), expect a band near 21 kDa. Include appropriate positive controls (e.g., lysates from human, mouse, or rat cell lines) and validate in your sample system.
- In immunohistochemistry (IHC-P), consider optimizing antigen retrieval and validate staining specificity using known positive and negative tissues.
- For ELISA, optimize coating antigen and detection conditions. Peptide competition or positive control antibodies may assist in assay development.
- As a polyclonal antibody, lot-to-lot consistency should be validated, particularly for quantitative or regulated applications.
