• HPS4 is a component of the BLOC-3 (biogenesis of lysosome-related organelles complex-3) complex, which acts as a guanine nucleotide exchange factor (GEF) for the small GTPases RAB32 and RAB38.

• Mutations in the HPS4 gene cause Hermansky-Pudlak syndrome type 4, a disorder characterized by oculocutaneous albinism, bleeding tendency, and lysosomal storage defects.
