Biological Background: HP1BP3 Function and Localization
- HP1BP3 (Heterochromatin protein 1-binding protein 3), also known as HP1-BP74, is a component of heterochromatin that maintains heterochromatin integrity during G1/S progression and regulates the duration of G1 phase to critically influence cell proliferative capacity. Related references: PMID:24830416
- It mediates chromatin condensation during hypoxia, leading to increased tumor cell viability, radio-resistance, chemo-resistance and self-renewal. Related references: PMID:25100860
- HP1BP3 localizes to the nucleus and chromosomes, consistent with its role in chromatin organization.
- The protein contains DNA-binding domains and repeats, and undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination (Ubl conjugation).
- Alternative splicing generates multiple isoforms; the full-length protein has a predicted molecular weight of approximately 61 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-160) of human HP1BP3, which is expected to recognize full-length HP1BP3 and possibly isoforms sharing this region. Consider sequence alignment if cross-reactivity with other HP1-binding proteins is a concern.
- For Western blot, the predicted molecular weight is ~61 kDa. Use a validated positive control lysate (e.g., HeLa or HEK293) and confirm specificity with siRNA knockdown or blocking peptide if needed.
- For IHC-P, test different antigen retrieval methods (e.g., citrate buffer pH 6.0) and antibody dilutions; validate in tissue samples known to express HP1BP3, such as tumor tissues.
- In ELISA, the polyclonal antibody can be used as a capture or detection reagent; for quantitative assays, consider optimizing the coating buffer and pairing with a matched monoclonal antibody.
- The antibody is reactive with human, mouse, and rat; however, performance may vary across species and applications, so validate in the relevant sample system.
