Biological Background: HP1 gamma/CBX3 Function and Localization
- Chromobox protein homolog 3, also known as HP1 gamma, HECH, or modifier 2 protein, is a conserved heterochromatin component encoded by the CBX3 gene.
- It recognizes and binds histone H3 tails methylated at Lys-9, leading to epigenetic repression. Related references: PMID:11242053
- It also binds histone H1.4 methylated at Lys-26, but is excluded from chromatin when H1.4 is simultaneously methylated at Lys-26 and phosphorylated at Ser-27. Related references: PMID:16127177
- HP1 gamma is involved in the formation of functional kinetochores through interaction with the MIS12 complex. Related references: PMID:15502821
- It contributes to the conversion of local chromatin to a heterochromatin-like repressive state via H3 Lys-9 trimethylation and mediates the recruitment of SUV39H1 and/or SUV39H2 by the PER complex to circadian target genes.
- The protein recruits NIPBL to sites of DNA double-strand breaks, playing a role in DNA damage response. Related references: PMID:28167679
- Subcellular localization is predominantly nuclear, consistent with its chromatin-associated functions.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin-like conjugation, which likely regulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-183 of human HP1 gamma, which encompasses the chromodomain and part of the chromoshadow domain; this design targets both functional domains.
- For Western blot (WB), the predicted molecular weight is 21 kDa; validate detection in your sample type and consider using nuclear extracts for enrichment.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization conditions to ensure epitope accessibility.
- The antibody is suitable for immunoprecipitation (IP) and ELISA; when using IP, confirm pull-down efficiency with appropriate controls.
- Cross-reactivity is reported for human, mouse, and rat; if using other species, verify sequence homology before testing.
