Biological Background: HP1 gamma/CBX3 Function and Localization

  • HP1 gamma, officially designated Chromobox protein homolog 3 (CBX3), is also known as HECH, HP1-GAMMA, and Modifier 2 protein.
  • It is a core component of heterochromatin that specifically recognizes and binds histone H3 tails methylated at lysine 9 (H3K9me), leading to epigenetic repression. Related references: PMID:11242053
  • Additionally, HP1 gamma binds histone H1.4 methylated at lysine 26 (H1.4K26me) and is excluded from chromatin when H1.4 is simultaneously methylated at K26 and phosphorylated at serine 27. Related references: PMID:16127177
  • The protein contributes to kinetochore assembly and function through interactions with the MIS12 complex. Related references: PMID:15502821
  • HP1 gamma also mediates the recruitment of NIPBL to DNA double-strand break sites, facilitating DNA damage repair processes. Related references: PMID:28167679
  • It plays a role in circadian rhythms by recruiting SUV39H1/2 methyltransferases to E-box elements of circadian target genes via the PER complex.
  • Subcellularly, HP1 gamma is localized to the nucleus.
  • The protein is subject to multiple post-translational modifications including acetylation, phosphorylation, and ubiquitination, which modulate its activity and chromatin interactions.

Experimental Guidance and Technical Tips

  • For Western blotting (WB), a band corresponding to the expected molecular weight of ~21 kDa can be observed. Consider optimizing antibody dilution and detection conditions with your sample type.
  • For immunohistochemistry (IHC-P), nuclear staining is anticipated; appropriate antigen retrieval and blocking steps should be optimized for each tissue type.
  • In ELISA, this antibody may be used as a capture or detection reagent for HP1 gamma/CBX3 quantification; assay conditions need to be standardized.
  • The immunogen spans the N-terminal region (aa 1–183) of the human protein, which shares high homology with mouse and rat orthologues, supporting reported cross-reactivity.
  • As this is a polyclonal antibody, batch-to-batch variability should be considered; validate performance in your relevant experimental systems.
