Biological Background: HOXA9 Function and Localization
- HOXA9 is a homeobox-containing transcription factor (alternative names: HOX1G, ABD-B-like) that provides cells with specific positional identities along the anterior-posterior axis during development.
- It acts as a sequence-specific DNA-binding protein that regulates gene expression programs critical for embryonic patterning and hematopoiesis.
- In endothelial cells, HOXA9 is required for the induction of SELE (E-selectin) and VCAM1 at sites of inflammation. Related references: PMID:22269951
- HOXA9 positively regulates EIF4E-mediated mRNA nuclear export and enhances the translation efficiency of ODC mRNA by competing with EIF4E repressors such as PRH (by similarity).
- The protein localizes to both the nucleus and cytoplasm, consistent with its dual roles in transcriptional regulation and post-transcriptional control.
- HOXA9 contains a characteristic homeobox DNA-binding domain and is subject to methylation and chromosomal rearrangements; it is implicated as a proto-oncogene in certain leukemias.
Experimental Guidance and Technical Tips
- For Western blot, a band of approximately 30 kDa is expected; validate band specificity using positive and negative cell lysates.
- In immunohistochemistry (IHC-P), anticipate nuclear and/or cytoplasmic staining patterns; include appropriate tissue controls and optimize antigen retrieval conditions.
- When designing ELISA assays, consider pairing with a capture antibody targeting a different epitope; cross-reactivity with rat HOXA9 should be confirmed in relevant sample matrices.
- As the immunogen spans amino acids 173–272, the antibody may detect isoforms or fragments containing this region; confirm performance in the intended application with endogenous controls.
