Biological Background: HOPX Function and Localization

  • Encoded by the HOPX gene (aliases HOD, HOP, OB1, LAGY, TOTO, CAMEO, NECC1, SMAP31); produces an atypical homeodomain protein that does not bind DNA.
  • Functions as a transcriptional repressor by interacting with serum response factor (SRF) to modulate SRF-dependent cardiac-specific gene expression; overexpression leads to cardiac hypertrophy.
  • Acts as a tumor suppressor and is frequently down-regulated in cancers such as lung cancer, choriocarcinoma, and head and neck squamous cell carcinoma.
  • Serves as a co-chaperone for HSPA1A and HSPA1B, assisting in chaperone-mediated protein refolding.
