Biological Background: HOOK3 Function and Localization

  • Acts as an adapter protein linking the dynein motor complex to various cargos and converts dynein from a non-processive to a highly processive motor in the presence of dynactin. Predominantly recruits two dyneins, increasing both force and speed of the microtubule motor. Related references: PMID:25035494, PMID:33734450
  • Component of the FTS/Hook/FHIP (FHF) complex, which may promote vesicle trafficking and fusion via the HOPS complex and regulates clearance of endocytosed receptors such as MSR1. The FHF complex also promotes the distribution of the AP-4 complex to the perinuclear area of the cell. Related references: PMID:32073997
  • Participates in defining the architecture and localization of the Golgi complex.
  • Subcellular localization includes cytoplasm, cytoskeleton, and Golgi apparatus.
  • Post-translationally modified by acetylation and phosphorylation; contains coiled-coil domains; involved in microtubule-based protein transport.
  • (Microbial infection) May serve as a target for the Salmonella typhimurium spiC protein, which inactivates HOOK3, leading to a strong alteration in cellular trafficking.
