  • Homer1 is a postsynaptic density scaffolding protein that belongs to the Homer family and is encoded by the HOMER1 gene (alternative name SYN47). The protein exists in multiple isoforms produced by alternative splicing, including isoforms 1, 2, and 3, with distinct functional roles.
  • It binds and cross-links group I metabotropic glutamate receptors (GRM1, GRM5) with endoplasmic reticulum-associated inositol 1,4,5-trisphosphate receptors (ITPR1), thereby coupling surface receptor activation to intracellular calcium release.
  • Homer1 also links GRM1 to PI3 kinase through interaction with AGAP2, and forms high-order complexes with SHANK1, which is critical for dendritic spine structural and functional integrity.
  • Isoform 1 regulates the trafficking and surface expression of GRM5, whereas isoform 3 acts as a natural dominant negative, competing with isoform 1 to modulate synaptic metabotropic glutamate function during long-lasting neuronal plasticity and development.
