Biological Background: hnRNP Q/SYNCRIP Function and Localization
- HNRNP Q (also known as SYNCRIP) is a member of the heterogeneous nuclear ribonucleoprotein (hnRNP) family, functioning as an RNA-binding protein implicated in various aspects of mRNA metabolism, including processing, stability, and translation.
- Multiple isoforms exist: Isoform 1, 2, and 3 associate with pre-mRNA, splicing intermediates, and mature mRNA complexes; isoform 1 specifically binds AU-rich sequences in apoB mRNA and participates in the APOB mRNA editosome; isoform 3 may be involved in cytoplasmic vesicle-based mRNA transport via synaptotagmin interaction.
- It is a component of the CRD-mediated complex that stabilizes MYC mRNA, and participates in the GAIT complex mediating interferon-gamma-induced translational silencing of inflammatory mRNAs.
- Localizes to both the nucleus (nucleoplasm) and cytoplasm, including microsomes and the endoplasmic reticulum, reflecting its dual roles in nuclear RNA processing and cytoplasmic mRNA regulation.
- Ubiquitously expressed in human tissues, including heart, brain, pancreas, placenta, spleen, lung, liver, skeletal muscle, kidney, and others, consistent with its fundamental role in gene expression.
- Post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitination, which may regulate its activity, localization, or interactions.
- Possesses RNA-binding motifs (glycine- and tyrosine-rich regions) that preferentially interact with poly(A) and poly(U) sequences, facilitating its participation in various ribonucleoprotein complexes.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (amino acids 524–623) of human hnRNP Q/SYNCRIP. This region may be conserved across species, contributing to the reported cross-reactivity with mouse and rat; however, sequence alignment is recommended for non-human samples.
- For Western blot (WB), the predicted molecular weight is 70 kDa; consider appropriate gel percentage and transfer conditions. Validate detection in the target cell or tissue lysate under reducing conditions.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization protocols may influence staining, as the protein is present in both nuclear and cytoplasmic compartments. Include appropriate controls.
- ELISA applications can be explored for quantitative detection; consider pairing with a capture/detection antibody system if high sensitivity is needed.
