Biological Background: hnRNP K Function and Localization

  • hnRNP K (Heterogeneous nuclear ribonucleoprotein K), also known as Transformation up-regulated nuclear protein (TUNP), is a major pre-mRNA-binding protein with high affinity for cytidine-rich sequences. It plays a critical role in pre-mRNA metabolism, including alternative splicing and mRNA processing.
  • In the DNA damage response, hnRNP K acts as a dual regulator of p53/TP53 signaling, functioning as a sumoylation-dependent transcriptional coactivator for p21/CDKN1A and 14-3-3 sigma/SFN induction, and as a repressor via interaction with the long non-coding RNA lincRNA-p21 to promote apoptosis.
  • hnRNP K participates in a ribonucleoprotein complex containing ZNF827, HNRNPL, and the circular RNA circZNF827, which associates with chromatin and may negatively regulate neuronal differentiation genes. Related references: PMID:33174841
  • Localizes to the nucleus (nucleoplasm), cytoplasm, and cell projections such as podosomes, supporting its dynamic shuttling and diverse roles.
  • Undergoes various post-translational modifications including acetylation, methylation, phosphorylation, and UBL conjugation (sumoylation), which modulate its activity and interactions.
  • In addition to RNA binding, hnRNP K can bind poly(C) single-stranded DNA and is implicated in host-virus interactions and intellectual disability when mutated.

  • For Western blot (WB), a band near 51 kDa is expected; nuclear fractionation may enhance detection due to hnRNP K’s enrichment in the nucleus.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), assess expression patterns in positive control tissues or cell lines, and optimize antigen retrieval and permeabilization conditions.
